Fiocruz’s Network Technological Platforms.

The Genomic Platform RPT01A is located at the Oswaldo Cruz Institute – in Rio de Janeiro/RJ. It has a genetic analyzer of the Sanger type, an ABI 3730XL model from Applied Biosystems, which allows the analysis of sequences of genetic material fragments (PCR products and plasmids). The set of 50 cm capillaries (96-channel capillary) allows readings of up to 900 nucleotides (nt), with Q>20, and up to 1100 nt with Q>10, with runs of around 2h/plate. The equipment can be supplied with up to 16 plates. Samples must be sent in tubes or plates.

Fiocruz’s Network Technological Platforms.

The Bioassays for screening anti-tuberculosis drugs platform performs microbiological tests to detect activity of natural and synthetic compounds against the M. tuberculosis complex, non-tuberculosis mycobacteria and others bacterias, using the broth microdilution method with oxidation indicator (MTT), colony count (UFC), BACTEC MGIT 960 and early bactericidal activity. This platform also perform tests with resistent strains. In addition, cytotoxicity assays using the MTT method and bactericidal activity in macrophage are offered when a compound shows activity against the mycobacteria tested.

Genotyping/Fragment Analysis.

The Genomic Platform RPT01D is located at the Oswaldo Cruz Institute – in Rio de Janeiro/RJ. It has a ABI 3130XL genetic analyzer model from Applied Biosystems, which aims to provide through a series of DNA fragment analysis methodologies, genotyping and microsatellites. The set of 36 cm capillaries (16-channel capillary) allows readings of up to 700 nucleotides (nt), with runs of around 6h/plate. The equipment can be supplied with up to 2 plates. Samples must be sent in tubes or plates.

This platform includes metrology and calibaration of equipment and instruments.

RBC (Brazilian Calibration Network) accredited service for calibrating instruments such as:

  • Measuring mass (Scale),
  • Temperature (Temperature Meter with Thermocouple Sensor, Temperature Meter with Thermoresistive Sensor or Other Sensors and Liquid in Glass Thermometer) and
  • Volume (Dispensers , Microvolume, Glass Pycnometer and Laboratory Glassware).

Fiocruz Network Facilities

The Electron Microscopy Facility gives support at sample preparation, training on proper use of instruments related to sample preparation and image acquisition. The FEI Tecnai G2 Spirit Biotwin Transmission Electron Microscope has a resolution of 0.34 nm and operates at a voltage of 20 kV to 120 kV, with options for tungsten (W) or lanthanum hexaboride (LaB6) filament and magnification of up to 300,000x. Equipped with MegaView Olympus 2K and Eagle 4K CCD cameras, as well as FEI’s Tecnai Imaging & Analysis (TIA) image capture software. Enabled for analyzing the ultrastructure of samples in 2D. Also, the JSM-5600LV Scanning Electron Microscope is a conventional high and low vacuum microscope with tungsten (W) filament and equipped with secondary and backscattered electron detectors for topographic analysis of samples. The microscope has a resolution of 5 nm and an acceleration voltage of 0.5-30 kV.

Fiocruz Network Facilities

The Flow Cytometry facility offers multicolor cell analysis, including phenotype, cell viability, proliferation, cell cycle, cell activation and measurement of cytokines. FACSAria III is a digital flow cytometer and cell separator (sorting) with three lasers (ultraviolet -355 nm, blue – 488 nm, red – 633 nm). It has the capacity to analyze up to 11 parameters, including size and granularity. FACSCalibur™ is an analog flow cytometer with two lasers (blue – 488 nm, red – 633 nm). It has the capacity to analyze up to 6 parameters, including size and granularity.

Fiocruz Network Facilities

The Illumina MiSeq Next Generation Facility provides sequencing library preparation and/or analysis for RNAseq, genomic sequencing, metagenomics and amplicon applications in single-end and pair-end formats. MiSeq has throughput ranging from 540 Mb to 15 Gb and 25 million reads.
