Fiocruz’s Network Technological Platforms
Nanobiotechnology Platform offers services for preparing and analyzing lipid and/or polymeric nanoparticles utilizing microfluidics technology. The platform provides the fabrication of nanoparticles and thorough physicochemical characterization, including assessments of size, zeta potential, and concentration.

The platform provides extract preparation and deposit services, from which samples will be prepared or plated for screening, confirmation of activity and determination of IC50 in in vitro and in vivo biological assays made available through collaborations signed with partner research groups. To perform these services, this platform uses the following set of equipment: Exhaust hood, Analytical balance, Rotaevaporator, Vacuum Pump, Vortex Shaker, Plate shaker, Ultrasound Bath, Single and multichannel manual pipettor, Multichannel electronic pipettor, SpeedVac evaporator and TLS labeler. Additionally, this platform provides Training and Plate Reading services on the multi-user SpectraMax M5 Reader equipment.

The RPT11H sets-up bacterial assays against various strains of gram-positive and gram-negative bacteria considered Multidrug-resistant organisms. The bacterial tests include preliminary assays such as antibiogram, minimum inhibitory concentration, and bactericidal and bacteriostatic activity. The platform also performs viability and cytotoxicity assays against non-cancerous and cancerous cells of human and animal origin, as well as antimalarial assays against the Plasmodium falciparum strain. All assays are standardized for screening all types of substances, whether they are extracts, fractions, isolates, oils, nanoparticles, nanofibers, peptides, etc. This platform is located in FIOCRUZ – Amazonas.

Equipment: Micromodulyo SAVANT freeze dryer for lyophilizing extracts, especially aqueous ones; Refrigerated Sheiker incubator, for the extraction process; Exhaust Hood – pre-evaporation of extracts; and Rotaevaporator, for concentration of extracts. This platform is located in FIOCRUZ – Amazonas.

Equipment: FACSCanto™ II, with two lasers (blue – 488 nm, red – 633 nm), with capacity to analyze up to 8 parameters, including size and granularity.

Two types of sequencing technology.
The next-generation sequencing Platform RPT01O, located in Manaus/AM, at the Leonidas and Maria Deane Institute – Fiocruz Amazonia, provides the preparation and analysis of DNA/RNA libraries for various applications, such as genomic or amplicon sequencing, transcriptomics, and metagenomics. It has MiSeq equipment producing up to 15 Gb (gigabases) and 25 million sequences reads, as well as NextSeq, which can reach 120 Gb (gigabases) and 100 million sequences reads.

The Genomic Platform RPT01H is located at the Leonidas and Maria Deane Institute – Fiocruz Amazonia in Manaus/AM. Since 2011, it has had a technical manager and a coordinator. It has a genetic analyzer of the Sanger type, an ABI 3130 model from Applied Biosystems, which allows the analysis of sequences of genetic material fragments (PCR products and plasmids) of up to 96 reactions every 24 hours with a 4-channel capillary, generating between 500 to 900 base pairs. The RPT-09-G subunit provides the equipment StepOnePlus™ and Quant Studio 5 (Thermofisher). Both thermocyclers with a detection system that uses LED lighting to detect four or six color channels respectively and are calibrated to operate with fluorophores: VIC®/ JOE™, ROX™, FAM™/SYBR® Green, TAMRA™/NED, or similar. In this way, the following tests can be carried out: SNP genotyping, gene expression analysis, microRNA expression, detection of genes, viral load analysis, and other absolute or relative quantitative assays. The following services are available:
a) Receipt of ready-made qPCR reactions using the TaqMan® methodology or similar fluorophores: VIC®/ JOE™, ROX™, FAM™/SYBR® Green, TAMRA™/NED or similar).
b) Receipt of ready-made qPCR reactions using the SYBR® Green methodology or similar.
c) Technical-scientific advice to assay design and interpretation

The Flow Cytometry Platform offers immunophenotyping data reading and analysis services; quantification of intracellular cytokines and culture supernatants, plasma and serum; cell proliferation and cell separation assays or “cell sorting”. The Platform currently has two cytometers, the FACSymphony S6 (19 colors/21 parameters) and the LSRFortessa (11 colors/13 parameters).

The Platform also has a FACSymphony S6 accessory compatible with sorting into plates, slides and tubes, thus offering the isolation/cloning service of populations of eukaryotic and prokaryotic cells, from suspensions obtained from biological fluids, cell line cultures, dissociated bodies and others. It also provides DIVA and Flow Jo software for analyzing results.

Fiocruz’s Network Technological Platforms.

The IGM Bioinformatics Platform was created in May 2012 and provides bioinformatics services to assist in research carried out at the institutional level. The services offered cover the creation, maintenance and mining of biological databases; analysis of RNA/DNA and protein sequences; training of personnel to use available bioinformatics resources and support for the development of new bioinformatics tools.
