Fiocruz’s Network Technological Platforms.
The Fiocruz Rondônia Bioprospecting Platform offers 23 specialized services in the preparation, analysis, and purification of bioactives. Services include fractionation and isolation of biomolecules using different liquid chromatography techniques, characterization of the isoelectric point and determination of the molecular mass of protein samples, determination of the partial sequence of amino acid residues from the N-terminal region of purified proteins, and screening of in vitro molecular interactions between ligands of recombinant origin and analytes using the surface plasmon resonance technique, as well as determination of ligand/analyte interaction constants.

Fiocruz’s Network Technological Platforms.

The Flow Cytometry Platform (RPT08I) offers immunophenotyping acquisition and analysis services; quantification of intracellular cytokines and cell supernatants, cultures, plasma and serum; cellular dermatology assays. The Platform currently has four cytometers, the CytoFLEX S (17 cores/19 parameters), Accuri C6 Plus (4 cores/6 parameters), FACSCelesta (12 cores/14 parameters) and FACSCan (5 cores/7 parameters). The Platform makes FCAP Array and FlowJo software available to users for analyzing results.

Fiocruz Rondônia’s Real Time PCR Platform has high-performance equipment and offers the entire scientific community the possibility of carrying out qualitative and/or quantitative reactions, in addition to evaluating gene expression. The platform also offers guidance on handling equipment and interpreting results with the aim of training qualified labor in the region.

Fiocruz’s Network Technological Platforms.

The Seq Studio Genetic Analyzer capillary sequencer can sequence fragments of up to 800bp. The capillary sequencing platform offers the ready plate reading service, where the user sends their already sequenced and purified samples only for reading on the equipment (capillary electrophoresis), and also the complete sequencing reaction service, where the user sends only the primers and their purified samples and the platform performs the entire sequencing and reading reaction on the equipment.

The Fiocruz Rondônia Microscopy Platform features modern equipment for imaging materials or live biological samples, such as intravital, in vitro, or ex vivo microscopy with fresh or fixed and stained histological sections (including immunofluorescence). These can be analyzed using the available equipment: Nikon Eclipse Ti Confocal Microscope, Nikon Eclipse 80i Fluorescence Microscope, Nikon ECLIPSE Ni-E Brightfield Microscope, and Nikon SMZ18 Stereomicroscope.

Fiocruz’s Network Technological Platforms
In vitro antiparasitic tests of natural compounds/extracts against promastigote forms of Leishmania spp. and asexual forms of Plasmodium falciparum W2. Cytotoxicity testing is also offered with cells of the J774 lineage (murine macrophages) and HepG2 (derived from a human hepatoma). All of these techniques are based on the use of fluorescence or absorbance detectors in plates of 96 or more wells, allowing the acquisition, automated analysis and determination of growth inhibition for 50% of the parasite population (IC50) and determination of the cytotoxic concentration for 50 % of cells (CC50), after treatment of each concentration of tested compounds.

Fiocruz’s Network Technological Platforms.

The FACSymphony A1 anAnalyzing Cytometer can simultaneously detect 18 parameters, incluing cell size and complexity and 16 different fluorescence. It also has the ability to detect microparticle assays (90nm to 500nm). The equipment has four lasers: • Violet (405nm) Filters: 450/50, 525/50, 610/20, 670/30, 710/50 and 780/60 • Blue (488nm) Filters: 530/30 and 710/ 50 • Yellow-green (561nm) Filters: 586/15, 610/20, 670/30, 710/50 and 780/60 • Red (637nm) Filters: 670/30, 710/50 and 780/60.

The building’s technological flagship is its level 3 biosafety laboratory designed to perform experimental infections of insects or vertebrates in order to study their role as vector and/or reservoir and to understand the interactions between hosts and pathogenic, particularly for arboviruses.

With a total surface area of 100 m2, composed of five rooms as well as its own laundry, the laboratory is notably equipped with several microbiological safety stations as well as all the prescribed and necessary equipment.

Fiocruz’s Network Technological Platforms.

The Genomic Platform RPT01A is located at the Oswaldo Cruz Institute – in Rio de Janeiro/RJ. It has a genetic analyzer of the Sanger type, an ABI 3730XL model from Applied Biosystems, which allows the analysis of sequences of genetic material fragments (PCR products and plasmids). The set of 50 cm capillaries (96-channel capillary) allows readings of up to 900 nucleotides (nt), with Q>20, and up to 1100 nt with Q>10, with runs of around 2h/plate. The equipment can be supplied with up to 16 plates. Samples must be sent in tubes or plates.

Fiocruz’s Network Technological Platforms.

The Bioassays for screening anti-tuberculosis drugs platform performs microbiological tests to detect activity of natural and synthetic compounds against the M. tuberculosis complex, non-tuberculosis mycobacteria and others bacterias, using the broth microdilution method with oxidation indicator (MTT), colony count (UFC), BACTEC MGIT 960 and early bactericidal activity. This platform also perform tests with resistent strains. In addition, cytotoxicity assays using the MTT method and bactericidal activity in macrophage are offered when a compound shows activity against the mycobacteria tested.
